Activity‐Based Protein Profiling of Oxidases and Reductases

Abstract Activity‐based protein profiling (ABPP) as a proteomic tool allows measuring the activity of enzyme classes or distinct proteins in their cellular context, annotating function uncharacterized proteins, and investigating target profile small molecule inhibitors. While hydrolases other classes, which exhibit characteristic nucleophilic residue, can be addressed by electrophilic activity‐based probes, large class oxidoreductases has received much less attention ABPP. In this review state art ABPP oxidases is presented. The most frequent principle for oxidative activation probe leading to an group exemplified alkyne amine based warheads. Alternatively, rare cases probes address residues certain oxidases. Most recently, with NQO2 first example reductase been reported.
